NITheP Webinar: Epigenetic modifications in obesity and type 2 diabetes by Yvonne Böttcher

Abstract: Obesity is a major healthcare challenge worldwide. Although genome-wide association studies (GWAS) were tremendously successful in identifying novel genetic variants associating with obesity only a part of the total variability can be explained so far. Moreover, adipose tissue distribution, especially visceral obesity is accompanied by increased risk for further metabolic conditions such as type 2 diabetes (T2D), dyslipidemia and hypertension. A strong contribution of non-genetic factors such as changes in DNA methylation correlates with obesity and may provide a link to its related diseases. We and others have shown that omental visceral adipose tissue (OVAT) has distinct gene expression and epigenetic profiles compared to subcutaneous adipose tissue (SAT), contributing to metabolic dysfunction. Several studies have shown that weight-loss or exercise significantly alter methylation levels in skeletal muscle or adipose tissue. Although via genetic association studies and mediation analyses some data hint at that methylation might be a consequence of obesity, it is still under debate, whether epigenetic alterations are cause or consequence of T2D or metabolic disorders. The talk shall give a short overview about the research field and will end up with some open research questions that I hope to approach in a collaboration with quantum biologists.
