Two dimensional electronic spectroscopy (2DES) is a technique which is able to reveal quantum coherent energy transfer in photosynthetic complexes such in reaction centres. Measurement techniques and data processing tools are developed to study these high efficient processes in a variety of photosynthetic compounds. The aim of our research is to gain a better understanding of the properties of bacterial reaction centres by studying the degree of quantum coherent energy transfer in wild type and a series of mutants of the RC of Rhodobacter sphaeriodes.
The steps required to process raw data from a 2DES measurement into actual 2D spectra (absorption vs emission) are discussed using a test-measurement on a B820 dimers as an example. In this talk the focus will be primarily on the techniques required to process the raw experimental data from it’s entering into the ccd into the 2D spectra containing meaningful information. Scattering removal using a double chopper technique, time domain filtering of the data, and the phasing of the 2D-data will be discussed. Preliminary results of recent measurements on bacterial reaction centres on the Amsterdam 2DES setup will be discussed.
